Cell-based assays

Cell-based assays are not always easy. Individual cells can react differently to external stimuli and following the binding and/or internalisation of added ligands is not always easy.
The LigandTracer instruments of Ridgeview can follow a group of cells and measure the binding or internalisation of (radio)labelled ligands.

LigandTracer

LigandTracer offers a straightforward and robust method for real-time interaction measurement on live cells.

The use of live cells takes you one step closer to in vivo situations and is helpful for understanding how e.g. a drug behaves in a natural environment in terms of affinity, interaction rates, binding mechanisms, specificity, and biological processes. The highly flexible nature of the technology enables a large number of additional applications such as the measurement of cell-cell, protein-bacteria and protein-protein interactions.

Experiments are performed in disposable cell dishes and cells or liquid do not come in contact with any part of the instrument, resulting in a maintenance free system. LigandTracer can measure at room temperature, +4 °C, and +37 °C, making it possible to study the thermodynamics of interactions as well as cell behavior. The computer delivered with the instrument contains pre-installed software packages for assay setup (LigandTracer Control), support ( LigandTracer Learning), and evaluation ( TraceDrawer), all designed to be intuitive and meet the user’s needs.

LigandTracer models

LigandTracer models differ based on the label type they detect and are named after the colour of their lids.

LigandTracer Green monitors fluorescent signals and is compatible with MultiDishes 2×2, which enables two parallel experiments simultaneously. The interchangeable LED-based detector lets you adapt the instrument to the fluorophores commonly used in your lab.

Available fluorescent detector models in 2020 (visit the website for an updated list):

  • Blue (488 nm) - Green (535 nm), e.g. FITC and Alexa 488
  • Yellow (590 nm) - Red (632 nm), e.g. Rhodamine Red
  • Red (632 nm) - NIR (670 nm), e.g. Alexa Fluor 633

LigandTracer Yellow/White/Grey detects the signal from radiolabeled ligands. Their supreme stability allows measurements of up to several days. Besides the regular LigandTracer applications, these instruments are primarily useful for characterizing diagnostic imaging agents and drugs for targeted radionuclide therapy.

Available instrument models:

  • LigandTracer Yellow: High-energy gamma, e.g. 99mTc, 11C, 111In, 177Lu, 18F, 89Zr, 188Re, and 68Ga.
  • LigandTracer White: Beta emission, e.g. 18F, 14C, 35S, 11C.
  • LigandTracer Grey: Low-energy gamma, e.g. 125I.

LigandTracer - Applications

The key to LigandTracer is its flexibility: its straightforward technology enables a wide use with many applications to choose from.

As long as one binding partner (the target) can be immobilized and the other be labeled with a fluorophore or radioactive nuclide (the ligand), you are free to choose what these partners should be. The most common target is a receptor expressed on live adherent or anchored suspension cells, but bacteria, tissue sections, or adsorbed proteins can serve as targets too. The ligand is typically a protein or peptide, but can also be small synthetic drugs, cells (cell-cell interactions), virus or nanoparticles.
